Hannah Spencer secured 40.7% of the vote in the Gorton and Denton constituency, with Labour coming in third with 25% and Reform UK second on 28%.
Spencer says the Greens have defeated the parties backed by billionaire donors.
Spencer's win is the first time the Green Party has ever won a UK by-election.
