Efficacy of a Tetravalent Dengue Vaccine in Healthy Children and AdolescentsCitationNEJM, 2019; 381(21): 2009–2019論文の要約本試験は、デング熱が風土的に流行しているアジアおよびラテンアメリカ地域の4〜16歳の健康な児童・若年者を対象に、タケダ社開発の四価デングワクチン（TAK-003）の有効性と安全性を評価した第3相無作為化二重盲検プラセボ対照試験である。対象と方法約20,000名を対象に、ワクチン2回投与群とプラセボ群に2：1で無作為に割り付け。発熱などで受診した被験者には血清型特異的RT-PCRによるウイルス確認が行われた。主要結果プロトコル準拠解析において、全体の発症予防効果（VE）は約 80.2%（95% CI: 73.3–85.3）。入院例に対してはさらに高い 95.4% の保護効果。ベースラインでデング熱未曝露（seronegative）だった群でも 74.9% の高い予防効果が認められた。有害事象の発生率はワクチン群とプラセボ群で同程度（それぞれ約3%前後）で、安全性も良好であった。
