Police expected 200,000 demonstrators, but as the protest got underway, that estimate nearly doubled to become one of Toronto's largest protests in recent months.
It was part of a global day of action called by Reza Pahlavi, Iran's exiled crown prince, as similar rallies took place in major cities around the world.
The largely peaceful protest in Toronto caused major road closures, transit delays and gridlock.
Many protesters draped themselves in the flag Iran used before the Islamic Revolution in 1979.
Protesters could also be heard chanting King Reza Pahlavi.
Organizers say they want to raise awareness of the revolution and get non-Iranians involved and to show people in Iran they stand with them.

Kearney reminded the president that Canada alone paid for the construction of the Gordie Howe Bridge, $4 billion, and those costs will be recouped from tolls once the bridge is open.
Once it is paid for, the revenue will be split evenly.
Former Michigan Governor Rick Snyder also notes that Canada financed the entire bridge, and Michigan and the U.S.
will get their half-ownership with no investment.
He says a delay in opening the bridge would be felt much more on the U.S.
side.
And Carney says workers and steel from both countries were involved in the construction.
But the Trump administration is insisting on negotiations to make sure the U.S.
gets its fair share of any proceeds.

Cuban aviation officials say for the next month, jet fuel will not be available at nine airports across the island, including the International Airport in Havana.
Air Canada says while it is suspending regular flights, it will send planes to Cuba to pick up Canadian travellers who are already there.
