I've had the pleasure of talking with a lot of really smart people in the last fourteen hundred episodes of the show. And our guest today is at another level. And his name is Dr. Andrew Salzman. Welcome to the show. Thank you. Thanks very much. Great to be here. Uh it's uh an honor to have you on. Gosh, there's so many directions we could go. Uh one of them is you're The first guy to really figure out what to do about BRCA, this gene that's associated with aggressive breast cancer. And that's not even our topic today, but you're an innovator in that field.
So let's just talk for a minute about that, because I think a lot of our listeners, expect about 58%, are women. And what happened with BRCA? How'd you get into that and breast cancer?
Well, sort of through uh a backdoor, we had been really fascinated by DNA injury, a DNA damage, and how it relates to reperfusion injury, ischemia, shock, inflammation. And with that kind of a focus, we had started to work on the role of polyADP ribospolymerase or PARP, which is the major DNA repair enzyme. So we were quite excited about that. We were looking models of myocardial infarction and stroke and so forth. And then right around that moment This was in two thousand four to two thousand five. There was the realization that PARP inhibitors, and we had developed the most potent one at the time, uh, could play an important role in uh treating cancer, specifically in BRACA deficient cancer. So we were looking at women with ovarian cancer, breast cancer, as well as pancreatic cancer in both uh genders, and and it was dramatic.
The effects of blocking PARP in that population with that cell type seemed to be really exciting. So that's how I got into it. Wow. And you just went
forward from there. And You eventually ended up looking at NAD and creatine and things like that. Well that was Well, that
